Histochemical Localization of a Chimeric Gene (rolC-GUS) Expression in Zygotic Embryos of Transgenic Tobacco Plants

Histochemical localization of the expression pattern of a chimeric gene (rolC-GUS) in zygotic embryo development in tobacco plants was analysed. The results indicate that strong expression was localized mainly in the vascular cylinders of the cotyledons and central axis of the hypocotyl. Quantitative analysis indicated an increase of gene expression in embryos up to 20 d after pollination (DAP), but decreased at 30 DAP. Continuous increase of GUS activity was recorded up to 12 d after imbibition (DAI) in germinating seeds. The xylem cells were visualized following phloem differentiation in the cotyledons at 3 DAI.Copyright 1994, 1999 Academic Press
